The development of a new high resolution genetic map of the mouse genome will be used to identify new loci and characterize their participation in SV40-induced transformation. The major focus of the proposal will be to investigate the structure of SV40 genomes and their expression in mouse fibroblasts transformed by this virus. For this purpose, techniques will be developed to study restriction-cleavage patterns of SV40 genomes and to introduce them into different cell lines. These cloned genomes will be used to investigate the mechanisms of transformation and for the analysis of the expression of SV40 genes. In addition, gene cloning techniques will be used to isolate cellular genes activated by SV40. The final aim of this research is to establish a general scheme for the analysis of SV40-induced transformation.Q:
